MOTS-c
Mechanism · Dosing · Effects
MOTS‑c is a 16–amino-acid mitochondrial-derived peptide (MDP) that acts as a metabolic regulator. It primarily promotes insulin sensitivity, fat oxidation, and improved exercise capacity, while countering age-related metabolic decline. Preclinical studies in animals suggest broad benefits for metabolic health, organ protection, and physical performance, though no clinical trials in humans have been completed to date.
Mechanism (how it works)
-
Activates AMPK signaling pathways → enhances energy metabolism and glucose uptake.
-
Promotes fat oxidation and increases energy expenditure → supports reduction of visceral fat.
-
Modulates mitochondrial function → counters age-related metabolic decline and improves exercise capacity.
-
Supports organ and bone health in preclinical models → liver, heart, pancreatic islets, and osteoblast activity.
Route: Subcutaneous injection, once daily
Schedule: 5 days ON / 2 days OFF
Typical cycle: 4 weeks (then 2–4 weeks OFF)
Dose range: 5–10 mg per week
Reconstitution
Add 3.0 mL bacteriostatic water to a 10 mg vial
→ Final concentration: ~3.33 mg/mL
Syringe reference:
At 3.33 mg/mL
• 1 unit = 33 mcg
• 30 units = 1 mg
• 60 units = 2 mg
Weekly Split Protocol
Beginner (5 mg/week)
Schedule: Monday–Friday
• 1 mg daily
• 30 units per injection
• Weekend OFF
Advanced (10 mg/week)
Schedule: Monday–Friday
• 2 mg daily
• 60 units per injection
• Weekend OFF
Titration Example
Week 1
→ 1 mg daily (30 units)
Week 2+
→ Increase only if tolerated
→ Up to 2 mg daily (60 units)
Timing
Best when mimicking natural mitochondrial signaling:
✔ Morning (fasted)
✔ 30–60 min pre-cardio
✔ On rest days for metabolic activation
Effects & Benefits
-
Metabolic support: Improves insulin sensitivity, glucose metabolism, and fat oxidation (preclinical evidence).
-
Weight and body composition: Promotes visceral fat reduction and energy expenditure.
-
Exercise performance: Enhances endurance and physical capacity; counters age-related frailty in animal models.
-
Organ protection: Supports liver, heart, pancreatic, and bone health in preclinical studies.
-
Anti-aging potential: Modulates mitochondrial function and metabolic pathways associated with longevity.
Side Effects & Safety Notes
-
No adverse effects reported in preclinical studies; human tolerability remains unknown.
-
Use caution, monitor for reactions, and consult a healthcare professional before use.
-
Use new sterile syringes for each injection and dispose safely.
